Добрый день,
В общем случае, ответить чётко нельзя. Если это GNOME, то, да, надо
добавить файл в POTFILES.in как-то так:
[type: gettext/glade]somefile.ui
Следует обратить внимание на то, что некоторые разработчики делают
опечатку. Например, в гномовском network-manager-applet написано так:
[type: gettest/glade]src/connection-editor/ce-page-bluetooth.ui
Вместо "gettest", надо "gettext".
Ну, дальше надо ./configure и make update-po в подкаталоге /po.
Встречаются также тяжёлые случаи (Inkscape) или yelp-xsl, когда надо
сначала всё пересобрать, а потом уже make update-po.
Наверное, стоило бы указать, откуда проблемка, чтобы ответ получился
точнее...
