Что-то я прочитал доку по winelib, но так и не понял, реально с помощью
неё собрать плеер написанный с применением direct show, кто-нибудь в курсе?

Почитайте доку еще... Там четко описано какая подсистема на сколько
процентов реализована... Затем посмотрите в своеи исходники и
составьте список используемых вызовов API, затем посмотрите в
исходники wine и узнайте все ли они реализованы...
А то ваш вопрос звучит как: могу ли я переплыть реку, ьез указания
ваших навыков плавания и названия реки...
